๐ Antimicrobial peptidomimetics are synthetic compounds designed to mimic natural peptides, targeting bacterial cell membranes to prevent the development of bacterial resistance.
๐ก The study aims to assess the ability of antimicrobial peptidomimetics to prevent bacterial resistance towards two commonly used antibiotics, Gentamicin and Ciprofloxacin, in ๐๐ต๐ข๐ฑ๐ฉ๐บ๐ญ๐ฐ๐ค๐ฐ๐ค๐ค๐ถ๐ด ๐ข๐ฏ๐ฅ ๐๐ด๐ฆ๐ถ๐ฅ๐ฐ๐ฎ๐ฐ๐ฏ๐ข๐ด bacteria over time.
๐ The Minimum Inhibitory Concentration and Fractional Inhibitory Concentration of antibiotics and peptidomimetics were determined using methods such as the checkerboard microtiter broth dilution method.
๐ Bacteria were passaged daily in the presence of antibiotics or a combination of antibiotics and peptidomimetics at certain concentrations. The MICs and FICs were tested daily and compared after 10 and 30-day periods.
๐ The research identified that ๐๐ต๐ข๐ฑ๐ฉ๐บ๐ญ๐ฐ๐ค๐ฐ๐ค๐ค๐ถ๐ด ๐ข๐ฏ๐ฅ ๐๐ด๐ฆ๐ถ๐ฅ๐ฐ๐ฎ๐ฐ๐ฏ๐ข๐ด bacteria developed resistance towards Gentamicin and Ciprofloxacin antibiotics when repeatedly exposed to them under sub-inhibitory concentrations over 10 days.
๐ However, when these antibiotics were combined with peptidomimetics, such as Melimine and Mel4, the Minimum Inhibition Concentration did not change, indicating no development of resistance.
๐ The study also revealed that the use of a combination of Gentamicin and Ciprofloxacin resulted in a less significant increase in MICs compared to the use of a single antibiotic.
๐ The effect of peptidomimetics did not alter even after 30 days of testing, representing their sustained effectiveness over time.
๐ The was no cross-resistance, meaning the bacteria did not develop resistance against different types of antibiotics.
๐ The study concluded that combining antibiotics with antimicrobial peptidomimetics can mitigate the development of antibiotic resistance in both It demonstrated the potential of peptidomimetics as effective antimicrobial adjuvants to reduce the rate of antibiotic resistance development and improve the effectiveness of current antibiotics.
